This is a notebook within a series on Grieder's experimental work at other labs that she rotated to. The notebook begins with diagrams of probes of DNA sub clone and genomic blots and bar graphs. Grieder used the macronucleus and micronucleus of the fertilized nucleus. The experiment showed the macronucleus had many fissions and phenotypic assortment. Grieder diagrammed sequence-specific DNA Deletion in Tetrahymena thermophila. The sequence was compared to Tetrhymena, Paramecium, Oxytricha DNA sequences. Oxytricha was similar to Tetrahymena. Oxytricha also had a micro and macro nuclear DNA. Grieder graphed molecular weight of C4A2 probe and constructed rather vague maps of Telomere clones, which showcased isolated subclones and digests. Grieder conducted a group meeting. Grieder showed the group the Guanine and Cytosine strand. Grieder found that the strand loops around with no discernable end point. The end point did not have a terminal G strand as Grieder predicted. Grieder diagrammed the exconjugants, where each of the two cells that separate after conjugation had taken place. A member of a conjugating pair of protozoan ciliates, in the Tetrahymena genus, after separation and before subsequent mitotic division of each of the exoconjugants. Grieder described the presence of bands in exoconjugannts. Grieder described the band, progeny type, combination of exoconjugation. Grieder also diagrammed out the Tetrahymena telomere structure, with base pair and gaps. The subsequent document is a electropolation protocol document alongside a list of clones which allowed Grieder to graph cell growth. Grieder also documented a series of genetic schemes to isolate and preserve lethal mutations in certain chromosomes that lack the excising and amplification of the rDNA. The document also included steps and objectives as well as patterns of assortment in genetic material (phenotypic assortment of rDNA). The notebook concludes with a general structure of micronucleus C4A2, comparing six of them as well as a subsequent documents on the result of the comparisons.
